Why scientists are concerned about leaks at biolabs

The theory that Covid-19 might be the result of scientific experiments has thrown a spotlight on the work of the world's most secure biolabs. 

While the evidence linking SARS-CoV-2 to the Wuhan Institute of Virology in China is strictly circumstantial, a number of experts want tougher controls on such facilities over fears that accidental leaks could touch off the next pandemic. 

Here's what you should know. 

59 top facilities 

The Wuhan lab belongs to the most secure class, commonly referred to as biosafety level 4, or BSL4. 

These are built to work safely and securely with the most dangerous bacteria and viruses that can cause serious diseases for which there are no known treatment or vaccines. 

"There are HVAC filtration systems, so that the virus can't escape through exhaust; any waste water that leaves the facility is treated with either chemicals or high temperatures to make sure that there's nothing alive," Gregory Koblentz, director of the Biodefense Graduate Program at George Mason University, told AFP. 

The researchers themselves are highly trained and wear hazmat suits. 

There are 59 such facilities across the world, according to a report Koblentz co-authored that was released this week. 

"There are no binding international standards for safe, secure, and responsible work on pathogens," the report, called Mapping Maximum Biological Containment Labs Globally, said. 

Accidents do happen 

Accidents can happen, sometimes at the top tier facilities, and much more frequently at lower rung labs of which there are thousands. 

Human H1N1 virus -- the same flu that caused the 1918 pandemic -- leaked in 1977 in the Soviet Union and China and spread worldwide. 

In 2001, a mentally disturbed employee at a US biolab mailed out anthrax spores across the country, killing five people. 

Two Chinese researchers exposed to SARS in 2004 spread the disease to others, killing one. 

In 2014, a handful of smallpox vials were uncovered during an Food and Drug Administration office move. 

Lynn Klotz, a senior science fellow at the Center for Arms Control and Non-Proliferation, has been sounding the alarm for many years about the public safety threats posed by such facilities. 

"Human errors constitute over 70 percent of the errors in laboratories," he told AFP, adding that US researchers have to rely on data from Freedom of Information requests to learn of these incidents. 

'Gain of function' controversy 

There is disagreement between the US government, which funded bat coronavirus research in Wuhan, and some independent scientists, about whether this work was controversial "gain of function" (GOF) research. 

GOF research entails modifying pathogens to make them more transmissible, deadlier, or better able to evade treatment and vaccines -- all to learn how to fight them better. 

This field has long been contentious. Debate reached a fever pitch when two research teams in 2011 showed they could make bird flu transmissible between mammals. 

Harvard epidemiologist Marc Lipsitch told AFP he was concerned "that it would create a strain of virus that if it infected a laboratory worker could not just kill that laboratory worker... but also cause a pandemic." 

"The research is not required and does not contribute to the development of drugs or vaccines," added molecular biologist Richard Ebright of Rutgers University, one of the staunchest opponents of this kind of research. 

In 2014 the US government announced a pause in federal funding for such work, which gave way in 2017 to a framework that would consider each application on a case-by-case basis. 

But the process has been criticized as lacking transparency and credibility. 

As late as last year, a nonprofit received funding from the US on research to "predict spillover potential" of bat coronavirus to humans in Wuhan. 

Questioned by Congress this week, Francis Collins and Anthony Fauci of the National Institutes of Health denied this amounted to gain of function research, but Ebright said it clearly does. 

The path ahead 

None of this means that Covid-19 definitely leaked from a lab -- in fact there is no hard scientific evidence in favor of natural origin or lab accident scenario, said Ebright. 

But there are certain lines of circumstantial evidence in favor of the latter. For instance, Wuhan is around 1,000 miles north of bat caves that harbor the ancestor virus, well out of the animals' flight range. 

Scientists from Wuhan were however known to be carrying out routine trips to those caves to take samples. 

Alina Chan, a molecular biologist from the Broad Institute, said there were no signs of risky pathogen research dying down in the wake of the pandemic -- in fact "it's possibly expanded." 

Last year, Chan published research showing that, unlike SARS, SARS-CoV-2 was not evolving fast when it was first detected in humans -- another piece of circumstantial evidence that could point to lab origin. 

Chan considers herself a "fence-sitter" on the competing hypotheses, but does not favor banning risky research, fearing it would then go underground. 

One solution "might just be as simple as moving these research institutes out into extremely remote areas...where you have to quarantine for two weeks before we re-enter in human society," she said.

Theory that Coronavirus Escaped from a Lab Lacks Evidence

The pathogen appears to have come from wild animals, virologists say, and there are no signs of genetic manipulation in the SARS-CoV-2 genome.

Since the early days of the COVID-19 epidemic, theories have circulated about the origin of the novel coronavirus causing the disease, SARS-CoV-2. One prominent rumor is that it first escaped from a lab in Wuhan studying bat coronaviruses and then spread to the public. This theory has also evolved into claims that the virus was genetically engineered to be a bioweapon. But scientists say that while there’s not enough information to pinpoint where the virus came from, there is no evidence that it was created in a lab.

The lab-escape theory had been circulating on social media and various blogs for weeks, but gained considerable visibility in a New York Post article in late February. In it, Steven Mosher, a social scientist and the president of the Population Research Institute in Front Royal, Virginia, summarizes why he believes SARS-CoV-2 may have been accidentally spread by China’s National Biosafety Laboratory at the Wuhan Institute of Virology, where researchers have studied bat coronaviruses.

In the article, Mosher describes several lines of reasoning, namely, that the lab is less than 10 miles away from the seafood market where a cluster of COVID-19 cases was first discovered, and that after the 2003 SARS outbreak, the SARS-CoV virus escaped from virology labs multiple times in China. He also describes how Chinese virologist and bioweapons expert Major General Chen Wei went to the Wuhan Institute of Virology with military scientists in January to study thenew virus, which Mosher sees as a form of damage control.

“The circumstantial evidence surrounding it is pretty compelling. . . . The idea that the epicenter of this epidemic would be just a few miles from the Institute of Virology in Wuhan, which is where we know that dangerous pathogens are being kept and looked at as potential bioweapons, I think the odds against that are just astronomical,” Mosher tells The Scientist.

Dimitrios Paraskevis, an epidemiologist at the National and Kapodistrian University of Athens in Greece, tells The Scientist that while it’s not possible to rule out the idea of lab escape, he believes that it is unlikely. “Any person who works in a lab must follow very strict safety regulations. It sounds to me very extraordinary that something happened and nobody took care about such an accident,” he says. The World Health Organization updated SARS surveillance guidelines in 2004 after the lab-based outbreaks, urging labs to follow proper biosafety procedures, and China replaced the director of its Center for Disease Control and Prevention.

One problem leading to a lot of apprehension and speculation about the new coronavirus is that scientists “don’t know what the actual source of the virus was,” Anthony Fehr, a coronavirus researcher at the University of Kansas, tells The Scientist. Additionally, researchers don’t know if SARS-CoV-2 immediately started to spread in humans after a single transmission from an animal, or if it took multiple zoonotic events between an infected animal population and humans.

Despite the question mark around the exact source of the disease, it does appear to have originally come from wildlife, according to a team of international public health scientists who wrote a statement published in The Lancet. An analysis of SARS-CoV-2 by scientists at the Wuhan Institute of Virology suggests that the virus’s genome is 96 percent similar to a coronavirus found in bats.

Mosher agrees that animals were the likely origin. “That doesn’t mean [the virus] wasn’t collected, brought to the lab, and was being tested on in various ways, and escaped from the lab,” he says. Mosher also does not claim that China genetically engineered the virus. “I’m not saying this has been genetically engineered to be a bioweapon that’s escaped from the lab. . . . I’m just saying that [China is] collecting dangerous pathogens, [and] they have a history of letting them escape from the lab,” he says.

Transmission from an animal, with no lab experiment or genetic manipulation involved, fits best with what scientists know about how other coronaviruses have made the jump to humans. In the past, these viruses have spread through wild bats that infect another type of animal—an intermediate host—that then spreads it to humans. SARS-CoV, for example, was transmitted from bats to civets to humans, while camels were an intermediate host in MERS, according to Quanta. The civet version of SARS-CoV was 99.8 percent similar to the one found in humans—much more closely related than the bat and human varieties of SARS-CoV-2—so researchers believe the new coronavirus also infected another type of animal on its way from bats to humans. But they have not found a candidate so far, according to Nature.

This ability to move in between different animal hosts is a characteristic feature of coronaviruses, according to Paul McCray, a pulmonologist at the University of Iowa Carver College of Medicine whose lab studies coronaviruses. “It’s exactly what we’ve learned in studies of SARS in 2002 and 2003, and MERS in 2012. . . . So the concept that this is happening again should come as no surprise,” he says. “For people that work with these viruses, this is completely unsurprising. We don’t need to come up with farfetched theories when the genome sequences and the characteristics of these viruses support what we’re seeing.”

No signs of engineering in SARS-CoV-2 genome

In addition to the claim that a naturally evolved virus escaped from a lab by mistake, some conspiracy theories have posited that SARS-CoV-2 was genetically engineered. In fact, researchers throughout the world, including in the US and China, have conducted research involving the creation of experimentally engineered hybrid coronaviruses. But there is no evidence that SARS-CoV-2 was genetically engineered, says Paraskevis, whose genomic analysis of the new virus was reported as a preprint in January.

RNA viruses, which include coronaviruses, “accumulate mutations at a rate one million times faster than human DNA [does]. . . . It gives them the ability to survive against an immune response,” Paraskevis says. While the new coronavirus does have some genetic differences to other known viruses due to mutations, “there’s no evidence that this is the result of a human experiment,” he says, adding that if the virus were engineered, scientists would expect to see additional genetic material in its genome. For example, an early bioRxiv preprint on SARS-CoV-2 found HIV-like genetic sequences, but online commenters pointed out that “the findings were at most a coincidence” and that research has since been retracted, reports STAT.

As there are still many unknowns about SARS-CoV-2, researchers worldwide are trying to uncover as much as they can about the virus. Chinese researchers “released the genomic sequence incredibly rapidly online. . . . They were very public in sharing the most important first piece of information” about it, McCray tells The Scientist. “The fact that scientists all over the world had access to that genomic sequence” made a lot of early research possible, he says.

Veggie burgers gain in popularity, meat sales down

Meat substitutes are gaining in popularity with Dutch shoppers while meat sales are going down, broadcaster NOS reported on Wednesday. 

Figures from market researcher IRI show that the sale of vegetable-based meat substitutes went up 51% over the past two years while meat sales went down by 9%. 

Chicken proved the exception with an increase in sales of 2% but the first half of this year saw a 0.2% falling off signalling that it, too, is becoming a less attractive consumer option.

Supermarket leaders Albert Heijn and Jumbo supermarkets now stock 100 and 200 meat substitutes respectively, the broadcaster said. In total, €123m was spent on substitutes in the last year, some 4.5% of the total spend on meat products. 

Traditional meat product producers and butcher shops are – sometimes reluctantly- adopting the trend for less polluting food, NOS said. Meat substitutes are much less harmful for the environment than meat, with 1 kilo of beef, the worst offender, producing over 57.9 kilos of CO2 and a kilo of tofu coming in at only 5.8 kilos of CO2. 

‘You have no choice. But we butchers are an innovative lot so we will turn our hand to meat substitutes,’ butcher of 50 years Gerard Steltenpool told the broadcaster. 

In January, website Nu.nl said the Dutch are eating less meat, with two in five people saying they cut down on steak and burgers last year. Around one third of the 20,000 people polled said they ate meat every day, while one in 10 is vegetarian and 7% fully vegan. 

Industry 

The Dutch meat industry is also adapting. Last December, multinational Unilever said it is taking over the Dutch fake meat company The Vegetarian Butcher ‘in response to the growing trend among consumers to increasingly opt for vegetarian and vegan meals’. 

Brabant-based meat processing giant Vion said in March it is seriously considering moving into the market for meat replacement products and in the future half the turnover of Dutch meat producing giant Zwanenburg will be based on non-meat products. 

‘The takeover of the Unox factory is a big step towards this goal,’ a Zwanenburg spokesperson told NOS. ‘For Unox we are making vegetarian sausages, soups and sauces.’ Including the vegetarian products produced by subsidiary Kips, Zwanenburg, vegetarian food now accounts for 30% of total sales.

Dogs trained to offer support to troubled US veterans

Nesconset (United States) (AFP) - Michael Kidd, now 84 years old, fought in the Korean War. His young German shepherd Millie helps calm him down when things start to swirl, usually at night.

Harry Stolberg -- a 42-year-old former Marine who served in Bosnia, Liberia and Nigeria -- has a chocolate Labrador named Rocky who wakes him up from his troubled dreams.

And 31-year-old Phil Davanzo -- who carried the bodies of fallen comrades during a hostage rescue operation that went wrong off Somalia in 2011 -- hopes his Rottweiler puppy will soon be trained to support him during his panic attacks.

The three US veterans, who all live on New York's Long Island, suffer from post-traumatic stress disorder (PTSD) and have sought solace through pet therapy -- namely, a loyal dog to keep them company.

The shelter animals are either trained or being trained to help them through difficult times by Paws of War, an association funded entirely by private donations that then provides the service dogs free of charge.

The group will also train a veteran's dog if he or she already has one.

"The biggest thing is he wakes me up from nightmares," Stolberg says of three-year-old Rocky.

"He can open the door, come in my room, turn on the lights, take my blinders off me... and lick my hands so I wake up."

Rocky, whose 18 months of training were completed six months ago, also helps Stolberg navigate large crowds, which can be triggering.

"If I can't move, he will get me out of that crowd -- he will pick up on that. He will walk around me and look at it and if I don't respond, he will walk away from the crowd with me hooked up to him," he says.

Paws of War -- their acronym is a play on POW, used to signify prisoners of war -- has been active since 2014.

More than 100 dogs have been trained so far, and the therapeutic results for their masters have been significant.

Off the meds

Kidd, who suffers from severe PTSD, has been able to reduce his intake of medication thanks to Millie.

At night when things are tough, "she will come over to me, she will put her paw on my shoulder, on my chest, and just give me a big slobber," says Kidd, whose father was in Normandy on D-Day.

"That's just saying, 'I am here for you.'"

Stolberg used to need sleeping pills to get through the night, but not anymore, thanks to Rocky.

"Sleeping was my biggest problem. (...) Now I only have a nightmare once or twice a month, instead of every day," he explains.

"A lot of that is also because I know that when I go to sleep, he is in the room -- he is going to wake me up no matter what."

'Quite a process'

It takes 18 to 24 months to teach dogs what to do when a troubled veteran sends distress signals, according to Rebecca Stromski, a senior trainer for Paws of War whose husband served in Afghanistan and Kuwait.

"It's quite a process actually, creating a mutual respect and a connection between the service dude and the service dog," she says.

"Once the foundations are in place and the dog starts feeling if things are going well or not for the veterans, they start to do certain motions when the guys are fidgeting," Stromski explains.

"I can start and cue that behavior and use that as an alert."

In the face of seemingly interminable wars for US military personnel in Afghanistan and Iraq, deployments which began in 2001, veterans are experiencing PTSD symptoms on a regular basis.

Paws of War has more requests for service animals than it can fulfill, with 50 veterans on the waiting list, according to the group's co-founder Dori Scofield.

Both ends of the leash

After running an animal shelter for 30 years, Scofield launched Paws of War after being contacted by veterans returning home from Afghanistan and Iraq.

Those soldiers had become attached to dogs they had found in those countries, but were unable to bring them back home.

Through word of mouth recommendations, the association quickly became a top meeting place for the 75,000-strong veteran community on Long Island, one of the biggest in the United States.

"We get applications every day -- we can't keep up," Scofield says. "I can't train enough dogs fast enough."

She has opened satellite offices in Florida and in northern New York state. She has also launched a free mobile veterinary clinic where veterans can bring their companion animals.

Dogs who might have ended up put to sleep in shelters now have homes, and veterans are rediscovering "a reason to get up every day, get moving, get out," Scofield says.

"It has been just so awesome, helping both ends of the leash."

Scientists revive brain function in dead pigs

Paris (AFP) - Scientists have managed to restore cell function in the brains of pigs hours after they died, in a breakthrough Wednesday that experts said threw into question the very notion of what makes animals -- or even humans -- alive.

Authors of the US-based study said their research could one day be used to help treat victims of heart attacks and strokes and unravel the mysteries of brain trauma.

In human and large mammal brains, cells crucial for neural function begin to degrade as soon as blood supply is cut -- a process long considered to be irreversible.

But the findings of the trial, published in the journal Nature, show that the brains of pigs can have their blood flow and cell function restored even hours after death.

The team from the NIH BRAIN initiative, a federally funded US research programme, used the brains of 32 pigs that had been slaughtered for food and discarded, without blood or glucose flow, for four hours each.

Then, using a tissue support system that pumps a liquid designed to mimic blood through organs, they rehydrated the brains for a further six hours.

Some brains received the patented life-support solution and others were given a control substance.

Flickers of life

The results were astonishing: the brains that received the artificial blood had basic cell function restored. Their blood vessel structure was revived, and the team even observed some localised processes -- including in synapses and immune responses -- flickering back to life.

Those that received the control solution turned within hours to what researchers described as "something a bit like yoghurt".

"I was tremendously surprised," Nenad Sestan, a researcher at Yale University and lead study author told AFP.

"We were surprised how well the structure was preserved. We found that cell death is reduced, which is very exciting and promising.

"The main conceptual finding is this: cell death in the brain occurs across a longer time period than we previously thought," Sestan added.

The team stressed that they saw no "higher level functional activity" such as electric signalling associated with consciousness in the revived brains.

"That is a clinical sign that the brain is alive, and we never found it," said Sestan. "This is not a living brain, it is a cellularly active brain."

Had any such activity been detected, the team said they would have had to deliver anaesthetic to the brains to null any pain -- despite them having been dead for hours.

"If some activity shows up that indicated consciousness, we would have to stop the experiment," said Stephen Latham, from Yale's Centre of Bioethics.

"This would be unique: inducing consciousness in an organ that's not connected to any living being."

'Not alive, not dead'

The findings suggest that scientists may have underappreciated the brain's capacity for self-restoration after a patient is declared brain dead.

But for experts invited to comment on the study, it raised deeper philosophical and ethical questions.

Writing in a linked editorial, Nita Farahany, a professor of law and philosophy at Duke University, said that the study "throws into question long-standing assumptions over what makes an animal -- or a human -- alive."

She said researchers had inadvertently created an ethical grey area where the pigs used were "not alive, but not completely dead."

Dominic Wilkinson, professor of medical ethics at the University of Oxford, said that the study could have vital implications for future brain research.

"This research reminds us that 'death' is less an event, and more of a process that occurs over time," he said.

"Cells within the human organism may be alive for some period of time after the human person has died."

Zvonimir Vrselja, also at Yale School of Medicine's Department of Neuroscience, said the team planned to use their support system on dead brains for longer periods of time in order to see how the cells react.

"We could potentially get some insight into how these brains could be saved," he told AFP.

You will discover that medi-physics is going to literally speak to cellular structure and give it instructions, without one chemical involved. There's always a reaction to chemistry, isn't there? There's always a side effect. When you push one thing, something else reacts, doesn't it? How do you like it so far? Your most elegant chemical designs, the ones that are helping with the worst diseases on the planet, all have side effects - and some of those side effects are death! I say to you, how do you like it so far?

Does it really seem elegant to you? Or perhaps it's just a more sophisticated way of bleeding in the barbershop? Dear ones, that's how you're going to look at it someday! You're going to slap your heads and say, "Remember the day when we did everything with chemistry and drugs?"

Right now, these kinds of changes have already started in several areas. There are discoveries being made that are healing some of the most heart-breaking diseases you have. I reveal one, because it's not a secret. I will always give you information that is either being worked on or has been postulated through free choice on this planet. That is the guideline of channelling. We can't give it to you. You've got to develop it yourself. However, we can put you in an energy that has a faster discovery potential - that is to say, discoveries become more obvious.

Alzheimer's is heart breaking. There are millions on this planet who develop this condition, more all the time. You're living longer and a plaque-like material that literally obfuscates your memory within the brain is becoming more common. EU states destroy suspect Polish beef as scare spreads

Warsaw (AFP) - Several EU countries on Friday seized and destroyed suspect beef from a Polish slaughterhouse where allegedly sick cows were butchered, despite assurances from Warsaw that the meat did not pose a health risk.

Poland's chief veterinarian Pawel Niemczuk confirmed that 2.7 tonnes of the suspect beef was exported, while the European Commission said the meat was traced to 13 member states where it was being withdrawn and destroyed.

The scare recalls a 2013 scandal in which horsemeat was passed off as beef and used in ready-to-eat meals sold across Europe.

Brussels will send a team of auditors to Poland to assess the situation on the spot, said Anca Paduraru, a spokeswoman for the European Commission.

French authorities said 795 kilogrammes (1,750 pounds) of beef from the slaughterhouse -- which has now been closed -- had been imported.

More than 500 kilogrammes have already been seized and destroyed, said France's Directorate General of Food.

French Agriculture Minister Didier Guillaume said nine French companies had been "duped" into importing beef from the abattoir in Kalinowo, a village some 100 kilometres (60 miles) northeast of Warsaw.

Lame or diseased?

Portuguese authorities said they had destroyed 99 kilogrammes of suspect Polish beef, while Romania said it had eliminated 1,432 kilogrammes as a "safety measure".

Sweden's National Food Agency said that just under 100 kilogrammes of the total 239 kilogrammes of suspect Polish beef delivered to the country had already been consumed, while the rest has been seized.

The beef was also exported to the Czech Republic, Estonia, Finland, Germany, Hungary, Latvia, Lithuania, Slovakia and Spain, according to the European Commission.

Niemczuk said another seven tonnes were sold to about 20 outlets in Poland but insisted that tests found it did not pose a health risk.

Despite the assurances, Polish beef producers are reeling from the scandal with prices falling by around seven percent since an undercover TV report on the slaughter was aired on Saturday.

"All segments of the Polish (beef) market are worried," Jerzy Wierzbicki, head of the Polish PZPBM meat producers' association, told AFP on Friday.

Wierzbicki insisted that the cows shown in the report by the TVN24 commercial news channel were lame but not diseased.

"They were lame cows... such cattle should not be taken to a slaughterhouse but should rather be put down on the farm by a person who is qualified to do so," Wierzbicki said.

'Isolated case'

"This is an isolated case, concerning a single company, the concerns are unfounded," he said, conceding that the scandal could deal a blow to Polish beef producers who rely on exports.

A leading pork supplier in the European Union, Poland also produced some 415,000 tonnes of beef last year and nearly 90 percent of it was exported, Wierzbicki said.

Poland's Agriculture Minister Jan Krzysztof Ardanowski said on Friday that "pathologies" in the agri-food industry would be "cauterised with a hot iron".

"A police investigation is underway regarding the owners of the slaughterhouse and anyone involved in this process," he said, adding that there was "zero tolerance" for anything that could harm Polish exports.

The criminal probe was launched after TVN24 aired footage of apparently sick or lame cows being dragged across the floor and then butchered.

The TVN24 report said dealers bought lame or sick cattle at much lower prices. A journalist working undercover at the abattoir used a hidden camera to film the secret late night slaughter when veterinary authorities were unlikely to visit.

The European Commission has not commented on the quality of the meat in question, but issued a statement that raised concerns about the treatment of the sick animals, which it insisted could not be considered fit for human consumption.

"The practice of dragging animals unable to walk, as described (in the Polish report) is prohibited by European Union legislation on the protection of animals in slaughterhouses," it said.
